活动目的:

20世纪80年代重组哺乳动物细胞系建立以来,并且随着分子生物学技术的不断发展,细胞用于生物制品特别是疫苗的生产、检测等方面越来越多。大肠杆菌、酵母、昆虫细胞和哺乳动物细胞都可以用于蛋白的表达,但真核细胞较于原核表达系统,具有高质量的生物活性蛋白质,同时在操作及应用中可分为瞬时转染短暂表达和稳定转染持续表达。因此,在疫苗生产过程前,科研人员应结合病毒或细菌生物学及免疫学特性,克服真核细胞表达产量低、操作系统复杂等缺点,选择合适的细胞系、培养基、培养条件和培养技术,以确保疫苗或其他生物制品在安全性和有效性的同时,还能保证经济收益。现在适用于规模化培养的细胞包扩悬浮细胞、贴壁细胞核半贴壁细胞培养。有生物反应器、细胞工厂和微载体等等细胞培养技术。除了生产,与细胞相关制品的质量控制与保证,也是所有生物制品生产人员更关注的问题,这对于工艺设计、种子库的来源和建立、工艺流程的确认和验证、产品检定方法的统一、人员培训都有更加严格的要求。
